IgY-12 (removes 12 abundant proteins from seraum.plasma.)

The Seppro® Tip IgY-12 is based on avian antibody (IgY)-antigen interactions and optimized buffers for sample loading, washing, eluting and regenerating. It is specifically designed to remove twelve high-abundant proteins from human biological fluids such as serum, plasma, and cerebral spinal fluid (CSF). This technology enables removal of Albumin, IgG, a1-antitrypsin, IgA, IgM, Transferrin, Haptoglobin, a1-acid glycoprotein (Orosomucoid), a2-Macroglobulin, HDL (Apolipoprotein AI & AII), and Fibrinogen in a single step. The targeted high-abundant proteins are simultaneously removed by the immobilized specific IgYs when crude biological samples are passed through the tip. Selective immunodepletion provides an enriched pool of low-abundant proteins for downstream proteomics analysis. Specific removal of twelve high-abundant proteins depletes approximately 90% of total protein mass from human serum or plasma. The low-abundant proteins in the flow-through fractions can be studied. Removal of high-abundant proteins enables improved resolution and dynamic range for one-dimensional gel electrophoresis (1DGE), two-dimensional gel electrophoresis (2DGE) and liquid chromatography/mass spectrometry (LC/MS). The collected flow-through fractions may need to be concentrated dependent upon the downstream applications.

Magtration-His^TM pre-filled reagent cartridges are designed to be used with variety of Ni²⁺ and Co²⁺ magnetic beads. Users can purchase the magnetic beads from different vendors (Table 1). A suitable type and amount of the magnetic beads can be selected by users to achieve purification by automation with a high purification yield. The magnetic beads can be regenerated for reuse using the same cartridges that are used for the purification.

No magnetic particles included in the kit, but customer can buy and apply commercially available Ni or Co magnetic particles in reagent prefilled PSS cartridges.

·        GST-Tagged Protein Purification

The Magtration-GST ^TM Protein Purification Kit includes the protocols and reagents to purify GST-fusion proteins from bacterial cell lysates and to perform GST-pulldown experiments in the Magtration® 12GC robot. The purifications and pulldowns are performed by binding GST-fusion proteins to user-supplied glutathione-magnetic beads. The beads are then washed and separated in the pipette tip using patented magnetic-filtration (Magtration®) technology.

All of the GST-protein purification protocols and GST-pulldown protocols are performed automatically with up to 12 samples processed in parallel. This design facilitates the rapid screening of samples for protein expression with the purification protocols and for analysis of multiple protein-protein interactions with the pulldown protocols. This kit features open reagent cartridges and bottled reagents that provide a low-cost alternative to pre-filled cartridges. Each kit includes color-coded cartridge templates to make filling easy and error-free.
